When is the last rear diff oil change?
If you have been in water lately, you could have some water in diff.
Pull the plug and check for color and smell.
I would start there.
Check shock mounts, the spare tire is tight up in the frame.

Check the rear u-joints on the drive shaft.

What kind of noise, clunk, growl, or whine?
Only when you first put the transmission in drive or while driving?
If while driving, under load or no load?

Squeaking noise.
Under load and when coming to a stop.
Nothing in reverse.
I suspect my wheel bearing is the culprit.
I took the parking brake assembly off to confirm it wasn’t the brakes.
Since it’s directional and under load, I still wouldn’t rule out the possibility of the rear U-joint either.

I got someone to get under the truck for me, and we have pinpointed it to the rear u-joint or in the diff itself.
Pulling the cover off now to inspect.
Hopefully, I don’t get a shiny surprise.
The Fluid looked new and had no shiny bits.
I did see some bubbles on the ring gear.
Smells like good gear oil as well.
How many miles do you have on that diff fluid?
As AKBob stated, I wouldn’t rule out the U-joint.
